Vaccinia virus A28L
This part includes the coding sequence (cds) of A28L, a membrane protein of vaccinia virus (VACV). A28L plays a key role in enveloping viral particles, and participates in the fusion of the virus with the host cell membrane, which is the first step for the virus to enter the cell. It also plays a role in the maturation and release of the viral particle, aiding in the formation of the viral particle and the subsequent infection cycle. Since A28L is located at the upstream region ahead of A27L in vaccinia virus genome, we used A28L sequence as a homologous arm for the recombination to mutate A27L in our project.
The cds of A28L was cloned by PCR amplification from the genome of vaccinia virus (GenBank ID: JX489136.1).
In our project, the A28L DNA sequence was served as a homologous arm for recombination only. Thus, the A28L sequence was not necessarily to be complete cds. However, considering the potential usage of A28L in the future or by others, we included the complete cds of A28L in this part.
